Which island would be the best to stay on if you are planning a one night, romantic stay and could you also give me restaurant recommendations for that island that would be a nice romantic choice?

I think your best choice would be San Juan Island with an itinerary something like this.
Take an earlier ferry from Anacortes to Friday Harbor so that you arrive in the morning or before noon.

Do some shopping around town or take a drive around the island stopping at the most interesting places and beaches.  Packing some cheese, wine, smoked salmon, and grapes would make a great snack to enjoy while overlooking the October seas from South Beach.

Make some reservations for the Duck Soup Inn http://www.ducksoupinn.com/ .  Other restaurants in town may have a better view but none can match the food served here.  Of course there are other great options however the duck soup is a personal favorite of mine.

As far as accommodations go and I would just try to find something with a fireplace and a Jacuzzi tub.  Use these websites as a starting point: http://www.Northwestrooms.com , http://www.SanJuansites.comhttp://www.thesanjuans.com

accommodations on orcas Island

accommodations on San Juan Island

Lopez Island accommodations

ferry schedule

Are you planning on walking on the ferry or taking your car?  I recommend you take your car in October because the ferry lines will be shorter than in high season.

Any other questions?

I vote for Orcas Island.  We bought property there because we thought it was the prettiest.

Have a romantic dinner at the Inn at Ship Bay http://www.innatshipbay.com/  Our friends who live on the island claim it's the best.  Reservations required.  They have a beautiful setting on the water and really pretty gardens so go early so you have time to look around.  Nice place to stay too and not too far from Eastsound.

Another really nice place to stay is Spring Bay Inn http://www.springbayinn.com/ if you and your significant other are athletic and would like to take a morning kayak trip.  New and clean and right on the water but very isolated.

There are lots of highly rated B&B's on the island.  Blue Heron, Kangeroo House, Kingfish Inn, Old Trout are all on the water and at the top of the list at tripadvisor.com.

The Outlook Inn is right in Eastsound and is quite nice. Especially good if you don't bring your car tho you might want to eat at Christine's http://www.christinas.net/ instead of Spring Bay since you can walk to Christine's.  (There's a shuttle from the ferry landing so you can leave the car and save yourself the hassle of the ferry lines.)

The Turtleback Farm Inn is not on the water but is in the book "1000 Places to See Before You Die."

For a smaller budget try the Cascade Harbor Inn or perhaps the Blackberry Beach Cabins if you can get their waterfront cabin.  http://www.blackberrybeach.com/  I've stayed at both and they both have beautiful views.

Ron, thank you very much for your input.  I wish I knew Orcas Island better myself; I always appreciate your expertise on orcas Island.

Right now I'm trying to fix the problem with the message board...

Thank you so much for all the recommendations.  We are coming on the ferry by car.  I am from Colorado and have never been up your way.  I'm really looking forward to it.  What is the weather like in October?
